Residents from Ekuphuleni township near Kenton-on-Sea on Monday blocked the busy R72 coastal route which links East London and Port Elizabeth with burning tyres.
Police gave protesters 15 minutes to disperse before using teargas to disperse them.
At least one passing vehicle was reportedly stoned.
