Mashatile retained his position after securing 531 votes to Nomvula Mokonyane's 356 at the much-anticipated ANC elective conference in Tshwane at the weekend.
Mashatile served as premier after Cope deputy president Mbhazima Shilowa resigned from the ANC in 2008.
Hardly six months in office, President Jacob Zuma removed Mashatile to make way for Mokonyane. The ANC Youth League and ANC structures in the province responded negatively and vowed to disrupt her term.
Votes for Mashatile by branch delegates had been interpreted as a clear sign of defiance of Zuma's decision to remove him. Mashatile is Deputy Minister of Arts and Culture.
Mashatile's supporters see his re-election as a one-way ticket back to the premiership.
"We want him to become the premier. We hope that those who supported Mokonyane using the two-centre of power argument will live up to their principles and support his premiership," said a supporter.
