This match will be played at the Ruimsig Stadium, a City of Johannesburg sport facility that is the traditional home of track and field athletics.
Ruimsig, which has since been refurbished, is one of the stadiums that will be used as official training venues for the 2010 Fifa World Cup.
"The match against Australia gives us a great opportunity a week before our opening match of the World Cup," said American coach Bob Bradley.
Bradley has assembled a formidable team that includes big names like Landon Donovan, Oguchi Onyewu and Clint Dempsey.
"Australia demonstrated in 2006 that they are a team capable of competing at the World Cup, and a match against a quality opponent on South African soil at that point in our preparations will help our readiness for game against England," Bradley added.
The match against the Socceroos will come after the US team has wrapped-up domestic preparations for the World Cup.
The Americans have lined up two "Send-off Series" matches against the Czech Republic on May 25 and Turkey four days later.
The US will meet the Czechs at Rentschler Field in East Hartford, Connecticut, before traveling to Philadelphia to face Turkey at Lincoln Financial Field.
Meanwhile, the All Whites of New Zealand will play Chile in a training match here in South Africa on June 9, at a venue still to be confirmed.
The game takes place days before New Zealand's opening World Cup match against Slovakia, said All Whites coach Ricki Herbert.
The All Whites, who have qualified for the World Cup for the first time since 1982, will also play friendlies against Australia, Serbia and Slovenia before coming to South Africa.
