Mthalane, 27, holds the IBF flyweight title while Tete, 21, is a step away from challenging him. Mthalane has been involved in 27 fights, more than double those of Tete, who is ranked eighth.
But Tete, who remains undefeated after 12 outings, must first deal with 10th-ranked Filipino Paul Garcia in a title eliminator on February 26.
Promoter Branco Milenkovic will present that fight after reaching an agreement with his Filipino associates.
"I dug deep for Garcia to come here. I hope Tete will repay me with a convincing win," said Milenkovic, who will stage the fight at Wembley Arena.

Already pundits have made their feelings known. Dingaan Thobela and Jacob Matlala tipped Mthalane to win. Sihle Mnguni, a former manager of Tete, and Phillip Ndou gave Tete their vote.
Mthalane is trained and managed by Nick Durandt, who also honed Tete's skills before he joined Vuyani Bungu.
Tete is managed by Butityi Konki. Bungu, the gentle giant of local boxing, said their focus for now was on Garcia.
Durandt said whoever the mandatory challenger is will lose to Mthalane "because we plan to make a number of defences. But Tete will be easy meat for Mthalane, and he knows what I am talking about".
