The former Muvhango actress, who was travelling with an SABC newsreader, was nabbed in Parktown, Johannesburg, and taken to the Hillbrow police cells.
She was charged with reckless driving.
Masebe was not the only celebrity who fell victim to the Johannesburg Metro Police Department's poor planning for the 94.7 cycling challenge. Metro FM DJ Wilson B Nkosi nearly missed his morning radio slot after driving for two hours to try to get to work.
"It normally takes me 20 minutes to get to work, but after two hours I found myself back in my neighbourhood," Nkosi told his listeners.
The race started in the morning in Midrand, went through the CBD, the south of Johannesburg, Diepsloot and ended in Midrand.
Masebe was released on a warning after it emerged that the arresting officer's charges against her were vague.
"The arresting officer became angry when Masebe asked why she was being arrested," said a highly placed source who had been on the scene.
"No one could tell her the reason for the arrest and they initially claimed it was for reckless driving."
A woman who called herself Tebogo Masebe - apparently the actress' sister-in-law - answered Masebe's cellphone. She said Masebe was "too traumatised" to speak.
"She is sleeping now," Tebogo said.
JMPD spokesperson Wayne Minnaar said: "Masebe wanted to cross over into St Andrews Road.
"She said she asked officers to let her cross and that one of them was rude," he said.
"She turned her car diagonally and blocked cyclists."
He said two officers asked her to remove her car but Masebe refused.
An officer arrested her and charged her with reckless driving.
"This would not have happened had she cooperated," Minnaar said .
He said Masebe was out on warning and would appear in court soon.
