"The findings reflect neither a balanced view nor the reality of events," said Mampeule.
Neither he nor his attorney had seen a full report of the charges brought against him.
The minister's decision to try to get rid of Mampeule comes after an investigation into the activities of the post office management and board members.
A department of communications spokesman, Albi Modise, said the investigation into corporate governance and management were made by an auditing firm.
The findings of the firm were given to the minister this month.
Matsepe-Casaburri launched an investigation last November following the suspension of Mampeule for what was called "undermining the authority of the board".
Mampeule had laid a criminal charge against his predecessor, Maanda Manyatshe, over a tender dispute.
Mampeule alleged that Manyatshe and other staff members were behind the irregular appointment of Vision Design House to create a new image for the Post Office at a cost of R99million.
This resulted in Manyatshe's resignation as managing director of MTN, to clear his name.
Yesterday, Matsepe-Casaburri briefed the 15-member Post Office board on the outcome of the investigation, which has not been made public.
She told the board that she was calling a shareholders' meeting next month "to consider the removal" of Mampeule as a director.
The government is the Post Office's only shareholder.
Matsepe-Casaburri said she was proposing the removal of Mampeule because of a loss of confidence in him and a breakdown of trust between him and the board.
"I am not in a position to comment further," he said. "I intend to cooperate fully with the disciplinary process. I remain confident that a fair and independent judge will clear me of all allegations." - Sapa
