Mathebula stopped Mkhuseli Kondile in the second round while Nzimande flattened Oneboy Makhwelo in the fifth round.
Mathebula's fight with Kondile, a former South African and WBF International junior lightweight champion, was scheduled for 10 rounds.
Mathebula, the WBC International champion, who is not a renowned puncher, dropped Kondile twice in round two before Kondile, from Port Elizabeth, was saved from further punishment.
It was Mathebula's 10th knockout in 19 wins, with one loss and two draws, while Kondile suffered his ninth defeat against 23 wins (19 KOs) and a draw.
A businesslike Nzimande cemented his stay as the number one contender for the national junior lightweight title with a stunning knockout.
He improved to 13 wins (six KOs), two losses and two draws while Makhwelo, who was counted out while still grimacing in pain on the canvass, dropped to 12 losses against 14 victories (nine KOs) and a draw.
Defeat could have been detrimental to their promising careers. Victory has since put pressure on promoter Branco Milenkovic to deliver the big fights.
