Jobodwana, South Africa's biggest hope after his bronze in the 2015 World Championships, would clock the fastest time of the three, a 20.53, not enough to earn a fastest loser placing.
Leotlela would stop the clock at 20.59 while Munyai earned a 20.66.
Canada's Andre De Grasse was fastest with 20.09, while sprint King Usain Bolt and rival Justin Gatlin cantered across the line to win their heats and earn a place in the semi-finals, which take place on Thursday.
