The tournaments will also be used to test the readiness of the two facilities ahead of the 32-nation quadrennial soccer extravaganza.
In Polokwane, an international competition features Kaizer Chiefs, SuperSport United, Bidvest Wits and visiting Brondby from Denmark.
The first game, scheduled to start at 10am, will be between United and Brondby, with Chiefs playing Wits in the second match at noon. The final is at 5pm.
Chiefs' general manager Bobby Motaung said: "It's a great honour to open a World Cup stadium and also play in a tournament to honour the late ANC leader Peter Mokaba.
"We have been training hard and are going to Polokwane to win the tournament, the support we have in Limpopo is incredible and we cannot afford to disappoint our fans."
United's communications manager David "Mazolman" Skhosana said: "Limpopo has always been our happy hunting ground. There is no two ways, the title will be ours."
Wits director George Mogotsi said: "It's good to be back in Polokwane. We are also honoured to be in this tournament and will give it our best shot."
Polokwane executive mayor Thabo Makanyane said: "We are over the moon to have a world-class stadium in Polokwane and are anxious to host the World Cup."
Players who will be seen in action include Kaizer Motaung Jnr (Chiefs), Anthony Laffor (United), Tefu Mashamaite (Wits) and Martin Bernburg (Brondby).
l Ajax Cape Town will play against neighbours Santos in the new Cape Town Soccer Festival where players like Nathan Paulse (Ajax) and Erwin Isaacs (Santos) will be on display.
