Kamte is the first black South African golfer to win on the European tour and is one of the favourites to clinch this tournament, which is sponsored for R13,6million.
Denmark's Anders Hansen is also coming back to the city that will host the opening and closing 2010 Fifa World Cup matches to defend the title he won last year.
Other big names include 2007 winner Ariel Canete from Argentina, 2007 champion Richard Sterne of South Africa, Darren Clarke from Northern Ireland, Edoardo Molinari of Italy as well as England's David Howell.
Besides Kamte and Sterne, other South Africans who will take part include Charl Schwartzel, James Kingston, Louis Oosthuizen and Anton Haig.
Several up-and-coming South African golfers, including Sipho Sithole, Muswalo Nethunzwi and Emmanuel Masoliedza, have also qualified through a 2009 final playoff game.
The event, which is expected to attract about 200 golfers from around the globe, is the third European Tour Order of Merit tournament in South Africa, along with the Dunhill Championship and the South African Airways Open.
It is co-sanctioned to form part of the European and Sunshine tours.
City of Johannesburg executive mayor Amos Masondo said the tournament was a great platform to showcase their readiness for the World Cup.
Masondo said they were delighted to have attracted some of the top golfers in the world.
