He was speaking ahead of their crunch second-round game against Western Province (WP) in Durban on Saturday at 3pm.
Despite coming up against rapidly improving teams including WP, the Blue Bulls and Griquas, Plumtree sees his team winning a home semifinal.
Defending champions Sharks wrapped up their first round with an emphatic 41-6 win over Griquas at home at the weekend.
They are on top of the table with 27 points from seven games, three points clear of WP in second, Griquas in third and the Bulls in fourth.
All three teams are level on 24 points.
Plumtree said yesterday that stiff challenges lie ahead but "I am happy with the way things have shaped up and what we have achieved thus far.
"All the teams will be coming to us guns blazing . but we will meet them head-on.
"My boys are raring to win the coveted trophy for the second time in a row," he said.
This weekend's full fixtures: Friday: Leopards v Boland (Potchefstroom).
Saturday: Sharks v WP (Durban); Griquas v Cheetahs (Kimberley); Lions v Bulls (Ellis Park).
